چکیده
Abstract The aberrant expression of microRNA (miRNA) is closely associated with various pathological processes, such as the development gastric cancer. High-efficiency quantification miRNAs significant for diagnosis, prognosis, and treatment cancers. However, sensitive reliable detection miRNA remains a huge challenge. We depict here novel fluorescent approach label-free by exploiting designed scaffold to integrate catalytic hairpin assembly primer exchange reaction (PER). In this method, that constructed based on hybridization between two structure probes (H1 probe H2 probe), capable specifically recognizing target activating signal amplification, PER process transcribes numerous G-rich sequences induce ThT-based generation. Based efficient amplification strategy generation mode, method exhibits wide range 7 orders magnitude high repeatability (coefficient variation, 2.76%), implying proposed will be robust tool in early diagnosis disease. Objective: The annual incidence of cancer in the world is growing rapidly. The most important factor in the cure of cancers is their early diagnosis. miRNA, as a biomarker for early detection of cancer, has attracted a lot of attention. Methods: In this study, an electrochemical biosensor was developed to detect the amount of miR-106a, the biomarker of gastric cancer, by modifying a glass...
متن کاملSensitive and selective label-free alkaline phosphatase detection based on DNA hairpin probe.
Here, we present a label-free, simple and signal-on architecture for fluorescent alkaline phosphatase (ALP) biosensor utilizing an SYBR Green I (SG) assisted fluorescence amplification method. The strategy relies on the fact that ALP provides a significant barrier to lambda exonuclease (λ exo) activity by a dephosphorylating DNA hairpin probe (HP), and SG shows a considerable fluorescence inten...
متن کاملAn enzyme free electrochemical biosensor for sensitive detection of miRNA with a high discrimination factor by coupling the strand displacement reaction and catalytic hairpin assembly recycling.
An isothermal, enzyme free, ultra-specific and ultra-sensitive protocol for electrochemical detection of miRNAs is proposed based on the toehold-mediated strand displacement reaction (SDR) and non-enzymatic catalytic hairpin reaction (CHA) recycling. The SDR was first triggered only in the presence of target miRNA and this process also affects other miRNA interferences having similar target seq...

The negative strand of tobacco ringspot virus satellite RNA is a self-cleaving RNA. Its catalytic domain and substrate domain have been identified, and the catalytic domain has been named hairpin catalytic RNA. Here we report the construction of a plasmid containing a modified hairpin catalytic RNA sequence that can be transcribed In vitro. Because this plasmid has two specific restriction enzy...
